Various fluorescent ruthenium and iridium aromatic amine complexes have been known to bind to double stranded DNA(ds-DNA) and utilization of the property for DNA-chip technology has long been attempted. Among various methods of detecting complexes bound specifically to ds-DNA, electrochemiluminescence (ECL) has attracted much attention as a technique that affords low price and high-speed detection method. In this study, we have synthesized ruthenium(II) and iridium(II) complexes of bpy (2, 2’-bipyridine), phen(1,10-phenanthroline), and ppy (2-phenylpyridine) respectively with large N-aromatic diimine intercalators and investigated the DNA binding affinity by spectroscopic methods and the ECL behaviors by DNA modified ECL electrode. The results indicated that Λ-[Ru(bpy)2(tpphz)]2+ (tpphz : tetrapyrido[3,2-a:2,3’-c:3”, 2”-h:2’’’,3’’’-j]-phenazine) has high affinity and selectivity for recognition of ds-DNA compared to the Δ-form complex and the other similar ruthenium complexes. It was also revealed that [Ir(ppy)2(tpphz)]+ recognized ds-DNA a little more efficiently than the ruthenium complexes though the affinity of the iridium complex to DNA was smaller than those of the ruthenium ones.【査読有】
